Elucidating the Functions of Plant Mannanases in Fine-Tuning Cell Wall Structure

My research focuses on elucidating the biochemical functions of plant mannanases, specifically targeting the MAN7 gene in Arabidopsis thaliana. Mannans are crucial hemicellulosic components of plant cell walls with significant roles in agriculture, bioenergy, and sustainable biomaterials. My project explores how these enzymes modulate mannan polysaccharides and aims to enhance plant traits such as disease resistance, fruit texture, and heavy metal tolerance. By leveraging Pichia pastoris, a methylotrophic yeast, as a surrogate host, I investigate how mannan modifications can support global food security, optimize agricultural productivity, and contribute to innovative biomaterials and prebiotic developments for human and animal health.

As part of my project, I am spearheading the investigation of AtMAN7's role in mannan metabolism. I design and clone the wild-type and mutant variants of MAN7 gene using modular cloning techniques, transforming these constructs into Pichia pastoris for functional expression. I oversee the transformation process, including propagation in E. coli and electrocompetent yeast cells, followed by genotyping and culturing to induce transgene expression. My responsibilities extend to quantifying monosaccharide content via ion chromatography, conducting fluorescence microscopy for protein localization, and interpreting the molecular phenotypes.
